Stable cell line development requires technical expertise and an in-depth understanding of cell biology to ensure functional, stable expression suitable for downstream cell-based studies.

SB’s cell biologists are highly experienced in developing stable cell lines expressing targets such as ion channels, GPCRs, enzymes and antibodies and understand the importance of identifying the right expression conditions for each target.

We screen high numbers of clones at an early stage to identify the best lines for further development. Following clonal expansion, our cell biologists conduct in-depth analysis to validate clones using the most sophisticated techniques. At each stage of the selection process, SB offers you complete control over which clones to advance, ensuring your final cell line has the phenotype that matches your needs.

Ion channels

SB have successfully cloned and expressed a range of ion channels including the complete sodium ion channel panel (Nav 1.1 – 1.8). Large channels such as these require careful cloning and sequence analysis followed by rigorous validation using both membrane potential assay and manual patch clamp electrophysiology in order to produce functional channels which are not toxic to their host.

GPCRs

While many GPCR cell lines are available “off the shelf”, it is important to include disease-relevant mutants in your studies. SB has first-hand experience of the differences in binding affinity between wild-type GPCRs  and disease-relevant mutants / splice variants and would be happy to clone and develop specific mutants to complete your assays.

Enzymes

Our enzyme kinetics team routinely run a range of enzymatic assays against targets such as kinases, PDEs, HDACs, methyltransferases and proteases. As such, they are well equipped to validate enzyme-expressing stable cell lines to confirm their functionality.

Antibodies

SB has developed recombinant antibody-expressing cell lines for use as both research tools and discovery of potential biological therapeutics. Selection can be followed by successive rounds of amplification to achieve maximum antibody yield.
